Development Of Amino Acid Focused Metabolomics And Its Application

Metabolomics is effective for understanding the current phenotype of living organisms by measuring and analyzing a wide variety of metabolites. Focused metabolomics are highly practical because they can be used to analyze highly quantitative data by focusing on important endogenous metabolites. The author developed precolumn liquid chromatography tandem mass spectrometry (LC/MS/MS) analysis methods for amino acids that arc components of proteins and have already been known to have physiological significance. A method that uses novel precolumn derivative reagents greatly exceeds the sensitivity and analysis speed of the conventional methods. We also developed an automated precolumn derivatization amino acid analyzer using a liquid chromatography mass spectrometer (LC-MS) that simplified this method. For high accuracy and precision analysis of amino acid concentration in human blood, we validated all steps from blood collection to measurement using this device, and built a test procedure useful for studying lice amino acid metabolornics in human plasma. While calculating the reference range of plasma amino acid concentrations in healthy individuals, it was found that the balance of tic amino acid concentrations is useful for risk screening of diseases, including cancers, diabetes, myocardial infarction. The derivatized LC/MS/MS analysis method was also useful for dipeptide, tripeptide and D,framino acid analysis, and amino acid analysis in tissue by imaging mass spectrometer.
