Treatment With Proteinase Inhibitor From Enterolobium Contortisiliquum In Mice With Asthma-Copd

Abstract
Background: Patients with asthma associated with chronic obstructive pulmonary disease presented symptoms with poor response to gold standard treatment. Proteinase inhibitor from Enterolobium contortisiliquum (EcTI) has anti-inflammatory activities and may represent a potential treatment for lung diseases. Aims and Objectives: We investigate the mechanisms involved in the effect of EcTI treatment in a model of asthma associated to Chronic obstructive Pulmonary Disease (COPD). Methods: Male Balb/c mice were divided in groups (n=8): SAL, OVA, ELA, OVA+ELA and OVA+ELA-EcTI. At day 29, we evaluated: exhaled nitric oxide (eNO), resistance (Rrs) and elastance (Ers) of respiratory system and bronchoalveolar lavage fluid (BALF) total cells. Data was statistically assessed and p<0.05 was considered significant. Results: There was an increase in %Rrs and BALF total cells in OVA+ELA compared to OVA and ELA (p<0.05). Observed a decrease in %Ers in OVA+ELA and ELA compared to OVA (p<0.05). We did not observe any difference in %Ers in ELA and OVA+ELA. In OVA+ELA-EcTI(16.5±1.9ppb) there was a decreased in eNO compared to OVA+ELA(41.0±5.1ppb, p<0.05). The %Rrs of OVA+ELA+EcTI(186.77±39.55%) decreased compared to OVA+ELA(625.15±81.05%, p<0.05). The %Ers in OVA+ELA-EcTI (118.91±28.10%) group increased compared to OVA+ELA (36.43±4.8%, p<0.05). BALF total cells in OVA+ELA-EcTI (5.4±1.0 x104cells/mL) decreased compared to OVA+ELA (23.2±2.9x104cells/mL, p<0.05). Conclusions: Our results show that EcTI treatment plays an important role in controlling lung mechanics and inflammation of the experimental model of asthma associated to COPD.
