Identification Of Lipidomic Responses To Gcrv Infection In Ctenopharyngodon Idellus Kidney (Cik) Cells

Grass carp reovirus (GCRV) is a causing pathogen of grass carp hemorrhagic disease, whose outbreak often brings out a huge economic loss in aquaculture filed. Cellular lipids play crucial roles in many biological processes, such as virus replication, maturation and transmission. Presently, it remains to elucidate the roles of these lipids in the GCRV infection. In this study, Here, a lipidomic approach was conducted in Ctenopharyngodon idellus kidney cells to examine the lipid profiling changes upon GCRV infection. The results showed that a total of 97 lipids species including 85 up-regulated and 12 down-regulated were identified with statistical significances upon GCRV infection. Mainly, neutral triacylglycerols were accumulated in the virus infected cells. Metabolic pathway analysis showed that these lipid species were markedly enriched in some signaling and immune responses pathways such as regulation of autophagy. The formation of lipid droplets and the triggered autophagy were also detected with staining and Western blotting. This study would enhance the understanding of GCRV infection and development of novel defense strategies targeting lipid metabolism for viral hemorrhagic disease.
