Protective Effect Against Acute Myocardial Ischemia Induced by Rutin From Buckwheat Flowers and Leaves Preconditioning in Rats

Abstract
Protective effects of pretreatment with rutin from buckwheat flowers and leaves (RBFL) were performed on a rat model of acute myocardial ischemia (AMI) and the model was established by ligating the left anterior descending (LAD) coronary artery for 6 h. Alteration of electrocardiogram (ECG) ST segment, myocardial infarct size, the level of lactate dehydrogenase (LDH), creatine kinase (CK), MB isoenzyme of creatine kinase (CK-MB), superoxide dismutase (SOD), malondialdehyde (MDA) and glutathione peroxidase (GSH-PX) were measured and myocardial tissue morphology was observed. Immunohistochemical technique was used to assay the protein expression of Bcl-2, Bax and Fas. RBFL (90 mg/kg/d and 180 mg/kg/d) preconditioning could inhibit the rising of ST segment, reduce the myocardial infarct size, decreases the LDH, CK, CK-MB and MDA level and increase SOD and GSH-PX level significantly in a dose-dependent manner. RBFL could also inhibit the Bax and Fas protein expression and promote Bcl-2 protein expression. The change of myocardial tissue pathological morphous was alleviated compared with the model group treated with RBFL. These findings suggest that RBFL preconditioning has the protective effects on AMI and the mechanism maybe related to modulate the protein expression of Bcl-2 family and Fas system.
