SIMPLE METHOD TO PROFILE ANTIOXIDANT-RELATED LEVELS OF AA, EPA AND DHA IN FAT-1 MICE

To investigate the antioxidant-related profile of omega-3 (eicosapentaenoic acid, EPA; docosahexaenoic acid, DHA) and omega-6 (arachidonic acid, AA) fatty acids in fat-1 transgenic mouse, we used a one-step gas chromatography (GC-FID) method to determine AA EPA and DHA in the tails of 6, 12, 28, and 38 weeks-old fat-1 mice. The calibration curves, sensitivity and accuracy of GC-FID were also calculated. The results indicated that the levels of EPA and DHA in fat-1 mice increased while AA and the AA/(EPA + DHA) ratio decreased significantly in an age-dependent manner compared to WT mice. Our study revealed an antioxidant-related dynamic profile of omega-3 and omega-6 in fat-1 transgenic mice and provided an overview to the conversion rate of antioxidant-related fatty acid metabolism.
