Abstract
目的 分析竹节参多糖不同浓度醇沉组分的单糖组成及相对含量.方法 采用乙醇分步沉淀法分离竹节参总多糖,得到4个多糖组分;各多糖组分经三氟乙酸水解、1-苯基-3-甲基-5-吡唑啉酮(1-pheny1-3-methy1-5-pyrazolone,PMP)柱前衍生化后,应用液相色谱-高分辨傅里叶变换离子回旋共振质谱(liquid chromatography coupled to high resolution fourier transform ion cyclotron resonance tandem mass spectrometry,LC-FT-ICR-MS)法和HPLC-紫外检测器法定性、定量分析各组分中的单糖种类和比例.结果 竹节参多糖各组分均由D-葡萄糖、L-鼠李糖、D-半乳糖醛酸、D-半乳糖和阿拉伯糖组成,其中以D-葡萄糖含量最高;随着醇沉浓度的升高,阿拉伯糖含量逐渐增加,而D-半乳糖醛酸含量逐渐降低.结论 PMP柱前衍生化HPLC法可用于竹节参多糖的单糖组成分析,不同浓度醇沉组分中单糖含量的差异可为竹节参多糖类成分的分离提供依据.
