坏死性凋亡在干细胞移植改善心搏骤停后神经功能的作用

Abstract
目的 探讨骨髓间充质干细胞(MSCs)移植对心搏骤停(CA)后大鼠脑组织受体相互作用蛋白激酶1和3(RIP1和RIP3)表达的影响.方法 该实验在福建省急诊医学研究所完成.SD大鼠随机(随机数字法)分为sham组(n=8)、CA组(n=8)和MSCs组(n=8).通过窒息法诱导CA后进行心肺复苏(CPR),复苏后1h,MSCs组和CA组分别经尾静脉注射1×106/0.5mL MSCs与等体积磷酸盐缓冲液.CPR后3d,对各组大鼠进行神经功能缺损评分(NDS),荧光显微镜下检测MSCs在脑内分布,脑组织HE染色观察皮层坏死神经元,Western blot检测脑组织RIP1和RIP3表达水平.组间比较采用方差分析或Kruskal-Wallis H检验.结果 CPR后3d,MSCs组NDS明显高于CA组[72.5(71.5,73.2)与63.0(62.5,64.1),Z=3.376,P=0.001],DAPI标记的MSCs主要分布在脑皮层,MSCs组皮层坏死神经元明显低于CA组[(29.6±5.9)%与(57.2±6.4)%,t=8.922,P<0.01],MSCs组脑组织RIP1和RIP3表达水平均低于CA组[RIP1:0.227(0.193,0.243)与0.599(0.535,0.629),Z=3.151,P=0.001;RIP3:0.217(0.203,0.274)与0.543(0.533,0.555),Z=3.361,P=0.001].结论 MSCs移植可能通过抑制坏死性凋亡从而改善CPR后大鼠神经功能.
