Expression of PUMA and its relationship with expression of Bel-2 and Bax protein in human glioma

Objective To study the expression of PUMA,Bcl-2 and Bax in human glioma cell and the relationship among PUMA,Bcl-2 and Bax proteins.Methods The expression of PUMA,Bcl-2 and Bax protein were examined by Western-blot analysis in 52 cases of human glioma cell and 10 cases of normal brain tissues.Results (1) There were significant differences in the expression of PUMA,Bcl-2 and Bax proteins between the control group and grade Ⅱ ,grade Ⅲ or grade Ⅳ groups(all P＜0.01).There was no significant difference in the expression of those proteins between the control and grade Ⅰ groups (P＞0.05).(2) There were correlations between the expression of PUMA,Bax or Bcl-2 proteins and tumor differentiation grade in human glioma (rPUMA=-0.925,rBax=-0.931,rBcl-2=0.945,all P＜0.001).(3) There were correlations between the expression of PUMA and Bax,Bcl-2 proteins in human glioma (r1=0.963,r2=-0.937,P＜0.001).Conclusion The expression of PUMA protein may be an important prognostic marker for human glioma.The expression of PUMA was correlated with tumor differentiation grade.So it could be used as a valuable marker for the prognosis of human glioma.\r\n\r\nKey words: \r\nGlioma;  Apoptosis;  PUMA;  Bcl-2;  Bax;  Western-blot
