Abstract
Objective To determine the role of PRL-3 in the proliferation of human SGC7901 gastric cancer cell line.Methods RNA interference,mediated by recombinant lentivirus expressing artificial PRL-3 miRNA,was employed to knockdown PRL-3 expression in human SGC7901 gastric cancer cells.MTT assay in vitro was conducted to determine the role of PRL-3 in the proliferation of SGC7901 cells.Results Recombinant lentivirus expressing artificial PRL-3 miRNA (Lenti.rPRL3-miRs)was constructed successfully.Transduction of PRL-3 miRNA lentivirus effectively silenced the PRL-3 expression in SGC7901 cells.Knockdown of PRL-3 significantly suppressed the proliferation of SGC7901 cells in vitro,and the transfected cells were decreased by 30.4% as compared with vacant vector group(P<0.01).Conclusion PRL-3 plays a key role in the growth of gastric cancer cell.and PRL-3 silencing inhibits gastric cancer cell growth.PRL-3 can be used as a potential therapeutic target to treat gastric cancer.\r\n\r\nKey words: \r\nStomach neoplasms:RNA interference:Lentivirus
