•Nrf2 is regulated by a number of biomolecules (e.g. Keap1, p62, PKC, NF-κB, miRNAs).•Decline of Nrf2 function has been observed in patients’ brains with Alzheimer’s disease (AD).•Gain of Nrf2 function shows therapeutic potentials in various AD models.•Lack of specific Nrf2 activators and limited suitable AD models are major challenges.
