Abstract
A plasmid pBlueBac-GM-CSF was constructed and cotransfected into Spodoptera fugiperda (Sf) cells with wild-type autographa californica nuclear polyhedrosis virus (AcNPV) DNA. The recombinant virus AcNPV-GM-CSF was identified and purified by four rounds of blue-plaque picked up. By assaying the culture medium, we demonstrated that the recombinant virus-infected Sf9 cells expressed hGM-CSF gene up to 2.7x10(5)CFU/ml Recombinant hGM-CSFs with apparent molecular masses of 15, 18 and 20kd were detected by the Western Blotting method.
