•Effective radiosensitizers for locally advanced pancreatic adenocarcinoma are limited.•DNA damage response inhibitors (DDRi) enhance radiosensitization in vitro.•Radiosensitizing effect is dependent of the type of DDRi and on the genomic backgrounds of the cells.•Ongoing clinical trials are investigating this combination in locally advanced pancreatic adenocarcinoma.
