•DLX-SLNs were prepared and optimized using nano-template engineering technique.•DLX-SLNs showed optimum physicochemical properties for efficient brain delivery.•DLX release from SLNs was prolonged for 24 h without initial burst release.•DLX-SLNs significantly reduced LPS-induced depressive behavior in rats.•Plasma and brain BDNF concentrations were significantly enhanced by DLX-SLNs.
