•MTX-AP-DCH Nps reach values <300 nm and always have a positive charge.•The encapsulation efficiency of MTX in the AP-DCH NPs is less than 60 %.•The release of MTX from the AP-DCH NPs is given by non-Fickian mechanisms.•MTX released from NPs enhanced the cytotoxic effect.
