•Retinoic acid and TGF-β1 is enough for IgA secretion but not for surface IgA in vitro.•The addition of rIL-5 and rIL-21 coordinately induce IgA+ cells in vitro.•IL-21R or IL-5R blockade in vivo led to lower frequencies of IgA+ in Peyer’s patches.
