Abstract
The design of oligopeptides, spatially complementary to blood protein-markers, such as myeloperoxidase (MPO) - a neutrophil enzyme which is a risk factor of myocardial infarction, stroke and other acute diseases, is an important direction in development of express-diagnostic tools, particularly biochips. To develop such tools the visualization and analysis of protein structures is needed, which was carried out using Protein 3D software developed earlier at ETU. In this program the conjugated ionic- hydrogen bonds systems (CIHBS) rendering is used in order to find complementary structures for sites of globular proteins. Using this approach the peptide sequences for determination of myeloperoxidase and its subunits in express - clinical and preclinical diagnostics of were developed. In order to optimize the interaction between MPO and peptide aptamer a series of samples were prepared which characterized with differing hydrophobic characteristics and UV absorbance, while preserving the same spatial characteristics.
