•Antrodia Cinnamomea is one of the fungus species and widely used as a medicine for cancer therapy.•We analyzed AC treatment on human liver cancer cells by proteomic strategy.•AC promotes misfolding of proteins and unbalancing of its redox status leading to ER-stress.
