地奥心血康对动脉粥样硬化大鼠TLR4/MyD88/NF-κB信号通路的调节作用

Abstract
观察地奥心血康对动脉粥样硬化大鼠TLR4/MyD88/NF-κB信号通路的影响,探讨其抗动脉粥样硬化的作用机制.将60只SD大鼠随机分为正常对照组,模型组,阿托伐他汀组(4.0 mg·kg-1),地奥心血康组(100,30,10 mg·kg-1),每组10只.采用高脂饲料加维生素D2复制动脉粥样硬化模型,第9周灌胃给药,连续8周.生化分析仪检测血脂TG,TC,LDL-C,HDL-C水平;ELISA法检测血清TNF-α,IL-6,IL-1β水平;主动脉苏丹Ⅳ和HE染色进行病理形态学观察;RT-PCR和Western blot法检测主动脉组织TLR4,MyD88,NF-κB p65 mRNA和蛋白表达.结果 显示,与模型组比较,阿托伐他汀组和地奥心血康高、中剂量组血清TC,TG,LDL-C含量显著降低,HDL-C含量显著升高,血清TNF-α,IL-6,IL-1β水平显著下降.阿托伐他汀组和地奥心血康组主动脉病变明显改善,主动脉TLR4,MyD88,NF-κB p65 mRNA和蛋白表达均下降.地奥心血康对大鼠动脉粥样硬化具有防治作用,其作用机制可能是通过调控TLR4/MyD88/NF-κB信号转导,减轻炎症反应,从而抑制动脉粥样硬化进程.
