•Histaminergic neurons of the tuberomammillary nucleus are excited by melanocortin receptor activation.•MC4Rs (but not MC3Rs) excite histaminergic neurons through a presynaptic glutamatergic mechanism.•MC4Rs in the perifornical lateral hypothalamus contribute toward melanocortin-induced activation of histaminergic neurons.•Chemogenetic inhibition of histaminergic neurons potentiates the anorexigenic effects of melanocortin agonism.
