•We characterized the expression and function of mospd2 in the barbel steed.•Mospd2 gene expression was altered in response to lipopolysaccharide treatment or Aeromonas hydrophila infection.•MOSPD2 protein is located on MO/MФ, neutrophil, and lymphocyte membranes.•MOSPD2 blockade significantly reduced monocytes/macrophages and neutrophils migration.
