How does diversity in the organization of secretory machines determine properties of neurotransmitter release? In this issue of Neuron, Rebola et al. (2019) found that distinct nanoscale assemblies of Ca channels and Munc13, not overall channel abundance, mediate differing release characteristics of two cerebellar synapses.
