Abstract 16841: Neutrophil-Mediated Docosahexaenoic Acid Oxidation Generates the Substrate for Integrin αMβ2 and αDβ2-Dependent Macrophage Migration

A protective role of DHA during cardiovascular diseases and inflammation has been recently questioned based on new data and clinical trials. The reported results demonstrate heterogeneous responses that can be attributed to the wide spectrum of DHA oxidation products with unknown functions. 2-(ω-carboxyethyl)pyrrole (CEP) is a product of DHA oxidation that forms adducts with proteins. CEP is generated predominantly during inflammation in macrophage-rich areas. The goal of this investigation was to evaluate the potential role and mechanism of CEP-related macrophage accumulation during inflammation. The role of CEP was assessed by testing leukocyte recruitment during thioglycollate-induced peritoneal inflammation. Anti-CEP antibody dramatically reduced macrophage accumulation in the inflamed peritoneal cavity, but had no effect on the early recruitment of neutrophils.
