•CoPcF16 and CMK-3 modified GCE was fabricated by a drop-removing method.•The electrode showed five pairs of well-defined redox peaks.•The combination of CoPcF16 and CMK-3 exhibited superior electrocatalytic activity.•Electrochemical discrimination of Cys from GSH was presented in biological samples.
