•Progestins activate mPRα on croaker and flounder sperm to induce hypermotility.•mPRα is coupled to a stimulatory G protein, Golf, on croaker and flounder sperm.•Progestins induce hypermotility via Acy/cAMP, Egfr/Mapk, and Pi3k/Akt/Pde pathways.•Progestin-induced hypermotility is mediated by an increase in sperm Ca2+ levels.
