•Structure-activity relationship of geranyl resorcylates derived from Hericium erinaceum was investigated.•Four natural products and five derivatives were synthesized.•Neuroprotective activities against endoplasmic reticulum (ER) stress-dependent cell death were evaluated.•Synthetic derivatives exhibited higher neuroprotective activities than natural products.
