•LacVax® OmpA elicited humoral and cellular immune responses in BALB/c mice.•Oral immunization with LacVax® OmpA elicited sIgA, a hallmark of mucosal immunity.•LacVax® OmpA immunized mice were protected against Shigella induced dysenteriae.•First report outlining protective potential of L. lactis based vaccine for Shigella.•IgG, sIgA, high levels of INF-γ and IL-10 correlated with protection from Shigella.
