•VLP coupling to microspheres is a viable approach for viral antibody detection in immunoassays.•VEEV IgG detection, this method was 2-log more sensitive than traditional plate-based ELISA, even with the same antigen.•Microspheres can accommodate multiple VLP architectures.•MAGPIX permits multiplexed detection of both IgG and IgM against VEEV and CHIKV humoral response in human clinical samples.
