HIV-1 Nef-GCC185 interaction regulates assembly of cellular protein complexes at TGN targeting MHC-I downregulation

A schematic model for MHC-I downregulation. The diagram representing an association of functional complex facilitating interaction of specific proteins for activation of signaling and stoichiometric pathway: Nef interacts with PACS-2 and transport Nef to GCC185, a TGN protein. The proteins associated with Nef are AP-1 and PI3K and with GCC185 are MHC-I and SFK which specifically interacts with each other at the functional complex. Nef interacts with SFK and recruits ZAP70 which in turn interacts with PI3K constituting the multi-kinase assembly, downregulating surface MHC-I at GCC185 on TGN. The Nef interaction with MHC-I induces structural changes in AP-1 resulting association of ternary complex for MHC-I lysosomal degradation.Unlabelled Image
