Abstract
该文探讨黄连提取物(extract of Coptidis Rhizoma,ECR)体外对白念珠菌丝侵袭的抑制作用.以XTT还原法评价ECR对白念珠菌丝代谢活性的影响;固体培养基上观测白念珠菌落边缘生长的情况;半固体培养基上观察ECR如何影响白念珠菌丝生长;扫描电镜观察ECR对白念珠菌丝形态的影响;荧光显微镜观察ECR对白念珠菌丝形态及活力的变化;qRT-PCR法检测白念珠菌丝侵袭素ALS3,SSA1基因的表达.结果 显示,XTF法检测到在64,128,256 mg·L-1ECR干预下,白念珠菌丝活性逐渐降低;128,256 mg·L-1 ECR在固体培养基上可抑制菌落褶皱,在半固体培养基中可明显抑制菌丝侵袭性生长;扫描电镜与荧光显微镜下发现,128,256 mg·L-1 ECR能显著抑制白念珠菌丝形成;qRT-PCR法显示,64,128,256 mg·L-1 ECR作用下,侵袭素ALS3,SSA1基因表达均下调,且在256 mg·L-1 ECR干预下,侵袭素ALS3,SSA1基因表达分别下调4.8,1.68倍.该研究表明,黄连提取物通过抑制菌丝生长及侵袭素表达从而影响白念珠菌的侵袭力.
