Abstract
目的 探讨葡萄糖、胰岛素对(G1RE)3BP-1启动子活性的影响及意义.方法 采用脂质体转染法将含有(G1RE)3BP-1启动子的真核表达质粒p(G1RE)3BP-1Luc转染至人肝癌细胞系-HepG-2细胞中,6 h后分别加入不同浓度的葡萄糖、胰岛素,孵育48 h后,分别检测其报告基因荧虫素酶的活性.结果 荧虫素酶的活性随着培养基中葡萄糖的浓度增高而增加,随着培养基中胰岛素浓度的增多而减少.结论 (G1RE)3BP-1启动子在肝细胞中受葡萄糖的正调控、受胰岛素的负调控,若用于调控胰岛素基因的表达,将使糖尿病的基因治疗更安全更可行.
