Effect of Daming capsule on the expression of potassium channel KV4.2 mRNA in myocardium of type 2 diabetic rats

OBJECTIVE: To study the effect of Daming capsule on the expression of potassium channel KV4.2 mRNA in the myocardium of the type 2 diabetic rats. METHODS: Type 2 diabetic rat model was established. The rats, whose plasma glucose were higher than 16.7 mmol·L-1, were divided into 4 groups randomly: high dose group(187.5 mg·kg-1·d -1), middle dose group(93.8 mg·kg-1·d -1), low dose group(46.9 mg·kg-1·d -1) and control group. The model rats were given with Daming capsule for 14 d. Plasma glucose was determined. The total RNA from the myocardium of rats was measured by reverse transcription polymerase chain reaction (RT-PCR). The effect of Daming capsule on the expression of potassium channel KV4.2 mRNA in the myocardium of the type 2 diabetic rats was observed. RESULTS: The expression of potassium channel KV4.2 mRNA in the myocardium of the type 2 diabetic rats was lower than that in the normal control group(P < 0.05), the expression of potassium channel KV4.2 mRNA in the myocardium was increased after being treated with Daming capsule(P < 0.05), the values of plasma glucose distinctly decreased. CONCLUSION: Daming capsule decreasd the plasma glucose, and increased the expression of potassium channel KV4.2 mRNA in the myocardium of the type 2 diabetic rats.
