•Chemical profile and biopotential of grapevine leaves were studied.•28 natural compounds were detected in leaf extracts using the LC-MS/MS analysis.•Autochthonous variety Sila yielded highest amounts of most of tested compounds.•Autochthonous variety Župljanka stood out with highest biopotential.•All leaf extracts were highly selective towards HeLa cell line.
