Abstract
Objective To investigate the effects of harmine on the expression of cyclooxygenase-2(COX-2), and the migration and invasion of a human gastric cancer cell line,MKN-45. Methods MKN-45cells were seeded in RPMI-1640medium supplemented with 10% heat-inactivated fetal calf serum and routinely incubated for 24h. After treatment with harmine at a final concentration of 2,4,8,16and 32μg/ml for 24,48and 72h,the cell proliferation was determined using MTT colorimetric assay.The expression of COX-2was detected by western blot analysis.In vitro wound-healing and transwell invasion assays were used to assess the effects of harmine on the migration and invasion of MKN-45cells. Results Harmine significantly suppressed the expression of COX-2in a dose-dependent manner(P0.01).Compared with control group,harmine significantly inhibited migration and invasion of MKN-45cells(P0.01). Conclusions This study demonstrates that harmine inhibits migration and invasion of human gastric cancer cells through down-regulating COX-2expression.
