Microrna-148a Improves Cell Biological Activities In Non-Small Cell Lung Cancer (Nsclc)

Aim: The aim of this study is to explain the effects and mechanisms of microRNA-148a in NSCLC. Methods: Collecting the adjacent normal and cancer tissues which were from the NSCLC patients. The Bim and miRNA-148 expressions were evaluated by IHC or ISH methods. The A549 cells were divided into 4 groups as Control, miRNA, miRNA inhibitor and miRNA inhibitor + Bim inhibitor groups. The cell proliferation and apoptosis were measured by MTT and flow cytometry methods. The cell invasion and migration abilities of difference groups were evaluated by transwell and wound healing assays. The relative proteins (Bim, Bcl-2 and EGFR) expressions were measured by WB assay. Results: The Bim protein and miRNA-148a expressions of tumor tissues were significantly differences compared with adjacent normal tissues (P < 0.05, respectively). Compared with Control group, the cell proliferation of miRNA inhibitor group was significantly suppressed (P < 0.01) and the cell apoptosis of miRNA inhibitor group was significantly up-regulation (P < 0.01); by the transwell and wound healing assays, the invasion cell number of miRNA inhibitor group was significantly depressed and the wound healing rate of miRNA inhibitor group was significantly down-regulation compared with that of Control group (P < 0.01, respectively). Compared with Control group, the Bim protein expression of miRNA inhibitor group was significantly stimulated and Bcl-2 and EGFR proteins expressions of miRNA inhibitor group were significantly depressed compared with those of Control group (P < 0.01, respectively). Conclusion: miRNA-148a down-regulation had effects to suppress A549 cell biological activities by targeting Bim in vitro study.
