•The scCO2/buffer system was firstly applied for the asymmetric hydrolysis of styrene oxide.•The scCO2/buffer system suppressed the non-enzymatic reaction and high ee value of (R)-PED was achieved.•The ee value of (R)-PED was further improved as the stepwise reaction was adopted.
