•Molecular dynamics is applied to study collision cascade process in graphite.•A deep insight into defects evolution and distribution is provided at atomic scale.•A hillock defect is repeated and two formation mechanisms are proposed.•Vacancy defect distribution in irradiated graphite is analyzed quantitatively.•An evidence of transform between different vacancy types is provided from a statistical view.
