•A convenient and sensitive CL sensing DNA are achieved by nanometer interface effect.•DNA detection is based on different nanometer interface effect after hybridization.•DNA action mechanism on nanometer interface is clarified to design other biosensors.•The sensitivity is due to direct signal amplification of nanometer interface effect.•(+)AuNPs’ strict distinction to different structure DNAs led to evident specificity.
