•This is the first report of direct regeneration protocol establishment from leaf cultures in H. perforatum plant.•Hypericum perforatum known for its anti-depressent properties is highly recalcitrant species for Agrobacterium tumefacience mediated genetic transformations.•A. tumefaciens mediated genetic transformation studies has been performed for the first time in H. perforatum plant which yielded gus gene containing transgenic plants.
