•Conformational change of myoglobin was explored in the presence of various ionic liquids in phosphate buffer by spectroscopic measurements.•1-Butyl-3-methyl cation can promote the unfolding of myoglobin.•Imidazolium cation with longer alkyl chain length has stronger denaturation ability for myoglobin.
