Real-time PCR (RT-PCR) assays using SYBR Green as an indicator of product amplification are thwarted by potential for poor specificity. We have validated a novel format for testing isolated colonies from plate cultures that is highly specific, uses 75% less reagents and circumvents preliminary extraction.
