•Selective tryptophan functionalization was applied in non-protected peptides with native sequence.•The model peptide modified through tryptophan late-stage lipidation conserved bioactivity.•Peptide glucagon with lipidation on tryptophan shown a greatly improved pharmacokinetic profile.
