•Mendelian randomization analysis revealed causal effect of alcohol.•Alcohol increases high-density lipoprotein (HDL) cholesterol levels irrespective of its lipoprotein size.•Alcohol decreases total low-density lipoprotein (LDL) cholesterol levels.•Alcohol might worsen LDL profiles by increasing a proportion of small-dense LDL.•Alcohol decreases triglyceride levels.
