P087 HLA-DPB1 allele typing frequency from a single center study

Abstract
Objective Limited by unavailable calculated panel reactive antibody (cPRA) for HLA-DP, it is impossible to predict the opportunity for a patient to receive a donor with compatible DP from a donor pool.The aim of this study is to provide a DPB1 allele typing frequency (TF) from a larger cohort as a reference for donor selection. Methods This study retrospectively reviewed 1354 patients and donors from solid organ and stem cell transplant registration at Cleveland Clinic who had been typed for HLA-DPB1 by high resolution sequence based typing.Four major US populations: Caucasian, African American, Hispanic, and Asian, along with mixed race were analyzed for DPB1 allele TF from total individuals and 234 normal related/unrelated donors. Results 35 DPB1 alleles were identified from this study.The total cohort analysis demonstrated that among the top 18 selected DPB1 alleles with the TF greater than 0.5%, HLA-DPB1 ∗ 04:01 was the most common allele in the population possessed by 69% of subjects, followed by ∗ 02:01 (27%), ∗ 04:02 (20%), ∗ 03:01 (19%), ∗ 01:01 (13%), which were present in subjects at TF greater than 10%.Sixteen alleles had high distribution TF in Caucasian.DPB1 ∗ 105:01 and ∗ 18:01 are the only alleles with highest TF in African American.Of the top 18 alleles, Asian subjects were found to have relatively higher TF with DPB1 ∗ 05:01 and ∗ 19:01 (Table 1a).Analysis of 234 normal individuals showed that DPB1 ∗ 04:01 was also found to be the most frequent (40%), followed by ∗ 02:01 (31%), ∗ 03:01 (18%), ∗ 04:02 (16%), ∗ 01:01 (9.8%), ∗ 05:01 (4.7%) (Table1b). Conclusion Our data provides DPB1 allele TF based on a larger cohort. The DPB1 allele TF calculated from the total cohort and from healthy donors had similar TF orders, which are in broad agreement with other published DPB1 allele frequency studies.
