Transcriptome Profiling of Retarded-Growth Rice Seedlings Induced by Low-Energy N+ Beam Implantation

The lack of transcriptome information about plants implanted by ion beam has retarded studies of ion-beam implantation/plant mutation breeding. Using RNA sequencing technology (RNA-Seq), transcriptomes of whole-rice seedlings generated from seeds implanted with a low-energy N+ beam were analysed 96 h after planting. We obtained 8.92 Gb short reads from three samples, with 8.05 Gb million high-quality 100-bp reads selected. A total of 776 transcripts were identified as DEGs in a sample of retarded-growth rice seedlings induced by N+ beam implantation: there were more than 544 DEGs in the sample of non-growth-retarded rice seedlings implanted by N+ beam. The analysis showed that there were more novel transcripts, completed novel alternative splicing events, and gene fusion events in rice exposed to ion beam implantation than in the control: these findings indicated the transcriptome complexity and various mechanisms of diversification by which the rice responded to ion beam implantation. This dataset provided a useful resource for plant ion-beam implantation research, and the use of this resource has enabled improvements to be made in the technology of ion-beam implantation into organisms and augmented future research in new areas of biology.
