Cloning and expression analysis of a C2H2 type zinc finger protein gene, JcZFP1,fromJatropha curcas L.

A novel zinc finger protein gene,JcZFP1,was cloned from Jatropha curcas L.endosperm cDNA library.The cDNA length was 931 bp which contained an open reading frame(ORF)of 735 bp,encoding aprotein of 244 amino acids.Its molecular weight and theoretical isoelectric point were 27.38 kDa and 7.0 respectively.The JcZFP1 protein contained a typical C2H2-type zinc finger domain including highly conserved amino acid sequences-QALGGH,which was a particular characteristic of plant ZFPs.Through Biology software prediction analysis,a lot of promoter-binding elements related to growth,development and abiotic stresses were found in the cis-acting elements of the gene.By fluorescence quantitative PCR,the expression levels of JcZFP1 were different as follows:green peelflower leafrootstem seed,which showed that JcZFP1 was expressed with high levels in the developing organs.Meanwhile,it was strongly induced by low temperature and drought in the Jatropha curcas L.seedlings.This indicated that JcZFP1 might play an important role in plant growth,development and response to environment stresses.
