Abstract
Objective: To investigate the effect of R-spondin1,a Wnt signal pathway activator,on osteogenic differentiation of human bone marrow mesenchymal stem cells( h BMSC). Methods:h BMSC were treated with 0,5,10,20 μg / L R-spondin1. Luciferase assay was employed to detect the effect of R-spondin1 on Wnt signal pathway,and Western blot was used to detect the expression of β-catenin which was the downstream target gene of Wnt signal pathway. Alkaline phosphatase( ALP) activity,calcium deposition,and the levels of OSX,OCN,RUNX2 were detected to observe the effect of R-spondin1 on h BMSC osteogenic differentiation. Results: R-spondin1 enhanced Wnt signal pathway,ALP activity,calcium deposition and the expression of OSX,OCN and RUNX2 in h BMSC osteogenic differentiation. Conclusion: R-spondin1 can enhance Wnt signal pathway of h BMSC and promote osteogenic differentiation.
