Abstract
目的:建立反相高效液相色谱法测定重组人胰岛素中胰岛素前体的方法.方法:采用Grace Vydac C18(250mm×4.6mm,5μm)色谱柱为分析柱,以乙腈-0.1％TFA水溶液(20:80)为流动相A,以乙腈-0.1％ TFA水溶液(80:20)为流动相B,梯度洗脱,流速为0.8 mL· min-1,检测波长为215 nm,柱温为40℃.结果:胰岛素前体在0.76～389.5μg· mL-1范围内峰面积与浓度有良好的线性关系(r=0.999 9),方法平均回收率为103.8％(n=9).结论:经方法学验证,本法能准确测定重组人胰岛素中胰岛素前体的含量.
