Abstract
With the demand of recombinant protein therapeutics changed constant,the expressing systems for therapeutic protein production should be improved. Recently,therapeutic proteins have been produced in microbial systems,insect cell systems and mammalian cell systems. Mammalian cells have become primary host cell for expressing recombinant protein. Among these,Chinese hamster ovary( CHO) cells are currently most widely used mammalian cells for the production of recombinant therapeutic proteins,because CHO cells can adapt suspension in serum-free medium for large culture and perform the complex post-translational modifications that are more stable and human-like. It is a key point to establish highly-efficient mammalian cell lines for recombinant protein production. To build high efficient mammalian cell line for expressing recombinant protein,many methods have been used to improve CHO cell line. People have focused on raising both cell-specific protein productivity( Qp) and the integrated viable cell density( IVCD). Recently,with the rapid development of biotechnology,mammalian cell expression systems for high production have been optimized. This review includes construction of expression vector,clone screening and cell engineering for high production Mammalian Cell line.
