柑橘类枯草杆菌蛋白酶基因CcSubtilisin的克隆及遗传转化

Abstract
以克里曼丁橘[Citrus clementina]叶片RNA为材料,采用RT-PCR技术克隆获得了一个编码类枯草杆菌蛋白酶的基因,命名为CcSubtilisin.该基因的开放阅读框(ORF)长度为2 337 bp,预测可编码778个氨基酸残基的多肽.分析发现CcSubtilisin与拟南芥、蓖麻、番茄、毛果杨、大豆、挪威云杉、欧洲桤木、百合等18个物种的同源蛋白具有相同的4个保守区域,属于枯草杆菌蛋白酶.通过构建该基因的超表达栽体并以沙田柚为材料进行遗传转化,获得了该基因的9个转基因沙田柚株系.实时定量PCR结果表明,4个用于检测的株系中有3株的表达量明显高于对照.
