Effects of Constraint-induced Movement Training on Promoting Myelinolysis Recovery in Rats with Ischemic Stroke

Abstract
Objective To study the effects of constraint-induced movement training( CIMT) on promoting myelinolysis recovery in rats with ischemic stroke. Methods 50 SD rats chosen and myelinolysis damgaged model of ischemic stroke prepared,and thet were randomly divided into CIMT group and nature recovery( NR) group. 10 days after operation,the CIMT group were trained with balance beam and rolling cage for 30 days,15 min/time,twice per day,with restricting the movement of the intact upper limbs. The NR group were trained with the same condition,but without restricting the movement of the intact upper limbs. Balance beam training scores were evaluated 15,20,25,40 and 60 days after operation of the two groups,and rolling cage training scores were evaluated 15,20 days after operation. Rats in both groups were checked with MRI and immolated for myelin staining. Results There was no statistically significant difference in balance beam training scores 15 days after operation between the two groups( P 0. 05),but balance beam training scores 20,25,40,60 days after operation in CIMT group were lower than those of NP group( P 0. 05). Rolling cage training scores in CIMT group were lower than those of NP group 15,20 days after operation. Myelin staining of the left cerebral peduncle: myelin staining was concentrated,within nucleated cells filled in NR group; a large of blue myelin staining and nucleated cells evenly distributed in CIMT group. Myelin staining of the right cerebral peduncle: cluster-like nerve tracts got thinner,myelinsheath lost,and nucleated cells reduced in NR group; nerve fibers of cerebral peduncle got thicker,some cluster-like nerve tracts distributed,and nucleated cells increased in CIMT group. The cerebral peduncle and brain pool in CIMT group did not changed as compared with the normal side,but the volume of cerebral peduncle got smaller and brain pool wider in NR group. Conclusion CIMT can promote the recovery of myelinolysis in rats with ischemic stroke,reduce the number of mylinolysis,and promote the motor function.
